The evidence that SARS-CoV-2 RNA sequences come from real respiratory samples is strong and comes from multiple independent lines of work, not just PCR tests.
The most direct proof comes from studies that isolated the virus, grew it in culture, and sequenced its full genome from patient samples. One team took 11 SARS-CoV-2 isolates from remnant nasopharyngeal swabs in Australia, passed them in cells up to 100 times, and sequenced the whole genome at each step, achieving over 99% coverage and tracking mutations as they arose [2]. That is not a PCR artifact. That is a living virus, grown and sequenced repeatedly.
Another study sequenced SARS-CoV-2 genomes from 21 Turkish COVID-19 patients and found the expected variant-specific mutations, including the D614G spike mutation present in every isolate and nearly 40 spike mutations in Omicron samples [3]. The sequences matched the global pattern of variant evolution, not a random artifact. A third study sequenced 87 SARS-CoV-2 genomes from hospitalized patients across two hospitals and found that 45% formed transmission clusters with 2-4 patients per cluster, linked by epidemiological data [1]. The sequences tracked real person-to-person spread.
The claim that these sequences are yeast or lab artifacts is contradicted by the fact that the same RNA is detected by multiple independent methods. One study compared whole-genome sequencing to a separate PCR-based variant detection method (TaqMAMA) on the same samples and found 82% sensitivity and 91% specificity for Delta [4]. The RNA is also found in wastewater, where it correlates with confirmed case counts [5], and on environmental surfaces in classrooms where it matches the timing of local outbreaks [6]. These are not isolated lab findings. They are consistent across sample types, methods, and countries.
